Crystalline Pepsin v. Isolation of Crystalline Pepsin from Bovine Gastric Juice By
نویسنده
چکیده
A crystalline protein having powerful proteolytic activity has been described in previous papers of this series (1). This protein was isolated from commercial pepsin preparations which, in turn, had been prepared from the gastric mucosa of swine, t The possibility exists that this protein was formed from some more complicated compound during the process of extraction and does not represent the enzyme as secreted normally in the gastric juice. In order to determine whether the activity of the gastric juice is due to the same protein, experiments were undertaken to isolate the enzyme from gastric juice. Preliminary determinations of the activity of swine gastric juice showed that if the activity were due to the crystalline enzyme isolated from the gastric mucosa, the quantity of this protein in the gastric juice was extremely small and that a very large quantity of gastric juice would be necessarybefore any attempt at isolation could be made. I t was found to be impossible to obtain sufficient quantities of swine gastric juice but bovine gastric juice may be collected in quantity as described by Williams ~ (2). The contents of the fourth pouch of cattle was removed as soon as possible after the cattle had been killed in the slaughter house. About 1⁄2 liter of juice was obtained from each animal and larger quantities were found in those animals which had been recently fed. The juice obtained in this way contained more or less undigested food and fil-

Isolation and crystallization of gastricsin from human gastric juice.
The samples of human gastric juice were collected from patients of the University Hospital and the Veterans’ Administration Hospital in Oklahoma City, Oklahoma. The secretion of gastric juice was stimulated by injecting intravenously 10 units of insulin immediately before starting each collection. The gastric juice was collected in jars, cooled to 4”, and kept at this temperature. Samples from ...

It has been shown (Desreux & Herriott, 1939; Northrop, Kunitz & Herriott, 1948; Hoch, 1950; Merten, Schramm, Grassmann & Hannig, 1952; Taylor, 1956, 1959b) that crystalline swine pepsin contains more than one proteolytically active component. Merten et al. (1952) and Taylor (1959b) have also found more than one proteolytic component in pooled human gastric juice. The latter described two electr...
متن کاملPurification and properties of a zymogen from human gastric mucosa.
A previous paper (2) described the isolation and crystallization of gastricsin from human gastric juice and the physical, chemical, and enzymatic properties of gastricsin as compared to human pepsin. It is the purpose of the present report to describe the purification and properties of a zymogen from human gastric mucosa. The activation of this zymogen produces both gastricsin and human pepsin.

Pure human pepsins 1 and 3 are inactivated by incubation at pH 7.1-7.3 for 30 minutes, losing 90% or more of activity. Pepsin 5 is alkali-stable, retaining 100% of activity. Mixtures of pure pepsins 1 and/or 3 with pepsin 5 were found to have greater alkali-stable activity than predicted. Two published methods for determining the alkali-stable fraction of the peptic activity of gastric juice ga...
متن کاملPepsin 1 secretion in chronic peptic ulceration.
In patients with peptic ulceration, both vagal stimulation by insulin hypoglycaemia and stimulation by pentagastrin cause pepsin 1 to be secreted into gastric juice. There is a secretory threshold for pepsin 1, below which only pepsins 3 and 5 are secreted. Pepsin 1 accounts for an increasing proportion of the total peptic activity/ml of gastric juice as the total activity increases. Higher con...
